Abstract

This study was conducted for the production of flavonoid from nodes. The main objectives of this study were to determine the effective disinfection method for P. minus Huds, to identify the best hormone and concentrations for callus induction and to determine the effect of hormones on concentrations of flavonoid content. This study was divided into two parts that was the in vitro production of callus using nodes, and then flavonoid extraction from callus using methanol extraction analyzed using HPLC. P. minus was obtained from villagers around Jeli and sterilized using 3 minutes of Tween 20, 100% bleach and 70% ethanol, then soaked with mercuric chloride for 5 minutes which can remove bacteria and fungus for 90%. Then the explants were cultured on MS basal media with concentration 1, 3 and 5 mg/L of four different auxins (NAA, IBA, IAA and 2,4-D) combined with 0.5 mg/L of BAP as control growth hormone applied in all concentrations media. The formation of calli, shoots and roots were observed. 5 mg/L of NAA recorded can induce callus at 3.60 ± 0.548 days faster than others, and the slowest nodes induced callus was 1 mg/L of IAA with 8.60 ± 0.548 days. The callus that produced highest flavonoid content was cultured in 1 mg/L of NM with 1.291552 mg/mL, and the lowest flavonoid content was callus cultured in 5 mg/L of IAA with 0.342392 mg/mL.
