Abstract

Meta-Topolin is a new cytokinin use for micropropagation. The effects of meta-Topolin (mT) on shoot and root regeneration of micropropagated “Musa acuminata cv. Berangan” was compared to benzyladeninepurine (BAP). Sterile shoot tip explants were cultured on Murashige and Skoog (MS) media containing 5, 10 and 15 mg/L of the mT and BAP. Two morphological growth of banana plantlets studied in this eksperiment which are the effect of mT on the shoot proliferation and the growth rate of in vitro
plantlets. After 3 subcultures, 5 mg/L mT led to the highest growth rate (the number of shoots and leaves and higher the shoots length) compared with BAP at the same
concentration. Plantlets were cultured on Murashige and Skoog (MS) media containing 5 mg/L of the mT. Since meta-Topolins is heat labile, it was added after the media being autoclaved and before media become solidified. Comparison between the effect of autoclaved and nonautoclaved met-Topolin was studied. Overall, the result obtained from this experiment do not give much difference in term of the morphology of the banana plantlets whether the mT being added into the media after being autoclave or non-autoclave. These current finding further demonstrate the potential of meta-topolin in banana micropropagation.
