The research was conducted to determine the effect of different media composition on the rooting of banana plantlets. The influence of different carbon source on the rooting of banana in vitro was compared by using sucrose, glucose, fructose, sorbitol, galactose and commercial sugar. The highest rate of roots per proliferated explant was measured based on the number of roots, root length and thickness produced. Significant length of roots and thickness of roots were obtained in the media containing commercial sugar as carbon source. Similarly, natural additives such as banana homogenate, potato extract and coconut water was tested to identify their effect on the rooting of banana plantlets. The best root growth was obtained in MS media containing banana extract as natural additives. Commercial sugar and banana homogenate are therefore can replace sucrose for rooting of banana in vitro to reduce the cost for media preparation. Finally, different minimal concentration of activated charcoal (Og/L, 0.5g/L, 1.0g/L, 2.0g/L) were used to determine the effect of activated charcoal on the banana rooting. Significant rooting was observed in banana culture on MS media supplemented with 0.5g/L activated charcoal.
