Abstract

This study was conducted to determine the effect of different sucrose and coconut water concentration on the micropropagation of Vanilla planifolia by using nodal stem as the explants. Results showed that explants cultured on MS media supplemented with 30g/L sucrose gave the highest shoot proliferation and growth with the mean number of shoot proliferation of 1.00 and the mean length of 2.00 cm, which is the best results compared to other sucrose concentrations. Similarly, in the study of effect of different concentration coconut water, the highest shoot proliferation and growth were also achieved by the explants cultured on MS media supplemented with 150 ml/L of coconut water with the same mean number of shoot proliferation and length. This indicated that the best combination of sucrose and coconut water on the MS media for efficient Vanilla planifolia micropropagation is the 30g/L sucrose + 150 ml/L coconut water together with 1 mg/L BAP. The findings of this study would help in large-scale in vitro mass propagation of Vanilla planifolia.
