Abstract

Java tea is (Orthosiphon stamineus Benth). To improve the quality of the plant, the chromosome number should be identified by karyotyping chromosome. Three methods of chromosome staining were tested on onion (Allium cepa) to get the best staining method. From the research, propionic orcein showed as the best staining solution by get the clearest image on root tips of onion (Allium cepa). Therefore, this method was used to karyotype java tea (Orthosiphon stamineus Benth. However, the metaphase stage of mitosis is unsuccessful to be seen because of the size of the chromosome is extremely small and might be suggested to be carried in future by using a different method.
