Abstract

Protein and protease from three selected germinated seedlings; Maize (Zea mays L.), Mung Beans (Vigna radiata L.) and Red Kidney Bean (Phaseolus vulgaris L.) was purified. The methods were partially purification using Ammonium Sulphate Precipitation and Dialysis. The protein and protease were purified using Ion Exchange Chromatography where DEAE-Cellulose acted as the resin and Size Exclusion Chromatography with Sephadex-G100 resin was used. The study was used Bradford's Protein Assay in the determination of the protein concentration after series of purification. Azocasein was used as substrate in assaying the activity of the protease. The study found Red Kidney Bean showed the highest protease activity with 34.667 U/ ml respectively followed by Maize and Mung Bean. Besides that, it also had the highest protein concentration compared to others after the purification using Size Exclusion Chromatography. The SDS PAGE analysis revealed that the molecular weight of Red Kidney Bean's Protein was 36 kDa, 48 kDa for Mung Bean and the estimation value for the Maize's protein sample was 40 kDa This result showed that Phaseolus vulgaris L. under study is a good producer of extra cellular protease, which can be beneficial for enzymatic industries and the consumers.
